Reprogramming the metabolism of Klebsiella pneumoniae for efficient 1,3-propanediol production

The production of 1,3-propanediol (1,3-PD) from glycerol by Klebsiella pneumoniae is limited synthesis numerous byproducts. Among them, the accumulation acetate has largest negative impact on fermentation performance. To address overflow caused knocking out lactate dehydrogenase, alcohol dehydrogenase and succinate several metabolic engineering manipulations were conducted. First, was reduced through enhancing assimilation pathway overexpressing heterologous acetyl-CoA synthetase. Then, polyhydroxybutyrate (PHB) introduced to further reprogram intracellular carbon metabolism. As a result, best performed strain Kpr-6 produced up 91.2 g/L extracellular 1,3-PD 2.56 PHB which can be easily separated each other, while dramatically reduced. strategies developed in this study would helpful for constructing microbial cell factory other similar bio-based chemical production.

Klebsiella pneumoniae LZ is a bacterium isolated from soil which can produce 1,3-propanediol from glycerol. Here we present a 5,431,750-bp assembly of its genome sequence. We annotated 9 coding sequences (CDSs) responsible for glycerol fermentation to 1,3-propanediol, 19 CDSs encoding glycerol utilization, and 134 CDSs related to its virulence and defense.
